研究室簡介

本實驗室主要應用分子生物學、細胞電生理及動態螢光測量技術來探討神經細胞的活性,特別是神經傳導機轉的調控及神經的死亡。
  1. 神經鈣離子感應蛋白 (Neuronal Ca2+ sensor proteins):鈣離子參與在許多細胞活動中。近年研究發現許多結構與鈣調素 (calmodulin) 類似的蛋白質,可以表現調控神經細胞的活性。我們已選殖出數個感應蛋白並確認與其相互作用的蛋白。當其表現在可興奮細胞中,會調節刺激分泌 - 耦合機轉的功能。
  2. 胞內鈣離子濃度恆定:胞內鈣離子濃度在休息態時,約為100 nM,當受到刺激時,會將濃度提升到數個uM。胞內鈣離子濃度的劇烈變化必須受到嚴格的調控,以儘速回到休息態。我們有興趣的是細胞膜上鈉鈣交換蛋白及胞內鈣庫如何調控胞內鈣離子濃度的恆定。
  3. 細胞胞吐與胞吞:當神經細胞興奮時,位於軸突末端的囊泡會融合至細胞膜,釋放貯存的神經傳導物質至突觸間隙。融合的細胞膜需要進行胞吞作用來維持膜的平衡穩定。我們有興趣的是囊泡運動,胞吐及回收相關機轉的調控因素。
  4. 鋅離子與神經退化:近年來鋅離子與神經退化疾病間的關係,已漸為清楚。我們有興趣於研究胞內鋅離子濃度變化的機轉,及與細胞自嗜機轉活化間的關聯,藉以了解鋅離子與細胞存活或凋亡間的關係。
  5. 奈米科技:透過與其他實驗室合作,我們使用奈米科技來偵測生物活性。近來我們成功的應用奈米線場效應電晶體來探討生物分子間的交互作用,並用以監測神經傳導物質的釋放;又如以原子力顯微鏡探究神經細胞表面性質,在各種刺激,甚或是氧化壓力下的變化。
